To detect the pesticide residues of organophosphate and carbamates in vegetables, fruits, grain, tea, water and soil.
Principle
Organophosphate and carbamate pesticides could inhibit the activity of acetylcholine enzyme existing in the insect central and peripheral nervous system, cause the accumulation of acetylcholine, and effect the normal conduction; so that make insect die from poisoning. The principle of this insect toxicology is used in the detection of pesticide residues. After adding reagent, we can determine the changing value of absorbance with time at 410nm using spectrophotometer, calculate the inhibition rate, and estimate the degree of organophosphate or carbamate pesticide residue in vegetables. Namely:
Substrate acetylthiocholine(AsCH) is hydrolyzed into thiocholine and acetic acid choline at the presence of acetylcholinesterase (AChE). The color reaction is produced based on thio-choline and 3-Carboxy-4-nitrophenyl disulfide (DTNB), leading to the reaction solution turns yellow which shows an absorption peak at 410 nm. Use spectrophotometer to estimate the inhibitory extent of organophosphate and carbamate pesticides on enzyme activity to know about the extent of pesticide residues ( To represent by inhibition rate) , thus determine the harmfulness.
